Agnogenic Myeloid Metaplasia – Causes and Evaluation

Causes of Agnogenic Myeloid Metaplasia

There is no concrete and definitive cause of Agnogenic Myeloid Metaplasia. Recent research indicates DNA mutations could be responsible for AMM. A specific mutation, JAK2V617F, is normally found in people with AMM. This mutation in JAK2 gene deregulates pathway signaling within your cells. As a result, your body now produces the wrong number of blood cells.

Other possible causes include:

– Mutations of other proteins in other pathways
– Deregulation of JAK pathway signaling
– Deregulation of mTOR and Ras pathways
– Gene activation
– Presence of leukemia and lymphoma

Your bones contain bone marrow, a soft fatty tissue. It consists of various immature stem cells. These produce blood cells. Your blood consists of red blood cells, white blood cells, and platelets. Red blood cells carry oxygen to different body parts, white blood cells fight infection, and platelets help in clotting of blood.

When genetic mutations scar bone marrow, production of blood cells is severely affected. This causes infections, bleeding, anemia, and other problems. Sometimes your liver or spleen may enlarge while trying to make blood cells. This is extramedullary hematopoiesis.

The reason behind genetic mutations is unknown. Mutated blood stem cells duplicate and divide. Mutations continue to occur in these new cells. As number of mutated cells increase, blood production in your body is disrupted. Your white blood cell count increases excessively and platelet count fluctuates.
